چکیده
Introduction The separation performance of the OptimaTM MAX ultracentrifuge is demonstrated by isolating a bacterial ribosome using two high-capacity rotors. This work is described in two separate application information bulletins: • Part 1 (document number A-1850A) describes conditions for pelleting an intact 70 S ribosome using differential centrifugation with a fixed angle MLA-80 rotor and an 8-mL tube. • Part 2 (this document) describes conditions for preparing and separating 30 S and 50 S ribosomal subunits using rate zonal density gradient centrifugation with a swinging bucket MLS-50 rotor and a 5-mL tube.

Ultracentrifugation in swinging-bucket and fixed-angle rotors evaluated for isolation and determination of high-density lipoprotein subfractions HDL2 and HDL3.
We evaluated the density-gradient ultracentrifugation method in a swinging-bucket rotor (Anal Biochem 111, 149-157, 1981) in a slightly modified version for isolation and determination of high-density lipoprotein (HDL) subfractions. We prestained the serum with Coomassie Brilliant Blue R, which did not change the hydrated densities of the lipoproteins, and after only 2.2 X 10(8) gav . min obtai...

BECKMAN stressing. ESP allows rotor speed to be reduced automatically before precipitation can occur. Since the time required for gradient formation decreases with faster rotor speeds, this approach results in more rapid separations. In general, for fixed angle rotors, rotor speed is limited by the need to avoid precipitation of the gradient salt at the bottom of the tube, where the concentrati...
متن کاملIsolation of hepatocellular lipid droplets: the separation of distinct subpopulations.
A new method for the isolation of lipid droplets from rat liver has been devised. The procedure involves tissue homogenization and discontinuous density gradient centrifugation. Six discrete bands of lipid particles, rich in triglyceride and cholesterol, are visible following 30 min of centrifugation at 25,770 g (max) in a swinging bucket rotor. An entire rat liver can be processed in 1-2 hr. D...
متن کاملPrecipitation with polyethylene glycol and density-gradient ultracentrifugation compared for determining high-density lipoprotein subclasses HDL2 and HDL3.
The purpose of this study was to compare quantification of cholesterol in high-density lipoprotein subfractions HDL2 and HDL3 by precipitation with polyethylene glycol (PEG) with that by density-gradient ultracentrifugation. Fresh serum samples from 32 fasting, obese children were analyzed with precipitation reagent "Quantolip" (Immuno AG), and then fractionated with a Beckman TL 100 ultracentr...
متن کاملA-1821A: Choosing Which Optical System of the OptimaTM XL-I Analytical Ultracentrifuge to Use
1 The Optima XL-I integrates both absorbance and interference optics on a single instrument; the Optima XL-A has absorbance optics only. 2 This optical system is called by a variety of names (Rayleigh optics, interference optics, and refractive optics). The names can be used interchangeably but it will be referred to here as Rayleigh interference optics. Choosing Which Optical System of the Opt...
